Abstract

EVALUATION OF ANTI-INFLAMMATORY ACTIVITY OF HERBAL FORMULATIONS OF DIFFERENT PROPORTIONS OF BRYOPHYLLUM PINNATUM AND PORTULACA OLERACEA

ABSTRACT

Background: Inflammation is a normal, protective response to tissue injury caused by physical trauma, noxious chemicals or microbiological agents. It is a complex reaction usually necrotic, cells that consist of vascular response, migration and activation of leucocytes and systemic reaction. Anti-inflammatory agents are use in the treatment of inflammation. There are two main types of anti-inflammatory agents; namely Glucocorticosteroids and Non- Steroidal Anti-inflammatory Drugs (NSAIDs). Bryophyllum pinnatum (Family: Crassulaceae) is emerging plant of interest of multipurpose use. It is commonly known as Patharchur. This plant has shown various pharmacological activities such as anti-helmentic, immunosuppressive, wound healing, hepatoprotective, antinociceptive, anti-inflammatory, antidiabetic, nephroprotective, antioxidant, antimicrobial, analgesic, anticonvulsant, and antipyretic activity. Portulaca oleracea L. (Family: Portulacaceae) is listed in the World Health Organization as one of the most used medicinal plants and it has been given the term „Global Panacea‟. It is commonly known as Kulfa saag. Portulaca oleracea possesses a wide spectrum of pharmacological properties such as neuroprotective, hepatoprotective, antidiabetic, antioxidant, antifatigue, anti-inflammatory and anticancer activities. Objective: Prolonged clinical use of NSAIDs used for treatment of inflammation is hampered by their side effects, most notably, gastric erosion, ulceration, haemorrhage, bronchospasm and kidney and liver dysfunction. Hence in order to avoid the unwanted effects while preventing the inflammatory process, our study emphasis to evaluate the anti-inflammatory activity of herbal formulation containing two different extracts of Bryophyllum pinnatum and portulaca oleracea in different proportions (1:1, 1:2 and 2:1) respectively. Methods: Dried leaves of Bryophyllum pinnatum and Portulaca oleracea were extracted with methanol (100% and 70%) by using soxhlet apparatus respectuively. Carrageenan and formalin-induced inflammation were used to evaluate anti-inflammatory activities of extract on acute and chronic inflammatory in wistar rat. Each experiment consisted of 25 animals randomly but equally divided into groups. Group A was taken as control and the Group E was treated with the standard drug Indomethacin (10 mg/kg). The methanol extract of herbal formulation of Bryophyllum pinnatum and Portulaca oleracea leaves 600 mg/kg were feed to group B, C and D in the proportion of 1:1,1:2 and 2:1 respectively. The data were analyzed using One way ANOVA, followed by the Post hoc Bonferroni Test and difference of means were considered significant at p<0.05 and p<0.01. Results: The herbal formulation has significantly decrease the development of paw edema induced by carrageenan and formalin in rats as compared to the control and standard drug group. The phytochemical screening of the methanolic extract of Bryophyllum pinnatum and Portulaca oleracea revealed the presence of flavonoids, tannins, saponins, triterpenes, cardiac glycosides, steroids, phenols and alkaloids. Conclusion: It has been concluded that the herbal formulation containing Bryophyllum pinnatum and Portulaca oleracea in ratio (1:1) exhibited better activity and higher potency than other two.
